I have a PDF fillable worksheet from my digital textbook that students complete in my Canvas course. Is there a way that it can be automatically graded like a Canvas quiz?

There is not a way to have PDF files auto-grade in Canvas. What you can do however, is create an assignment where students annotate the PDF file in Canvas. This way it makes it easy for students to complete and then it will show in the speedgrader.
