Does Canvas always indicate if a student has made changes to an assignment, such as a discussion post, prior to the due date?

Generally, I would that Canvas does a good job at showing student submission history for assignments, quizzes, and discussions. There may be some edge cases, especially if using 3rd party assignments, but they shouldn't be something you run into a lot. For discussions specifically, Canvas added an edit history to discussions recently. You can read the Adding Edit History to Discussion Redesign - Instructure Community - 586828 blog for more info on that if you'd like.
