Thank you for contacting the Instructure Community. It is possible the assignment was only setup for one submission. Teachers often keep assignments with only one submission attempt. Presuming that you have already uploaded once following this guide: How do I upload a file as an assignment submission... - Instructure Community and now can't upload a second time? You need to contact your teacher directly and ask them to allow a second attempt. Hopefully this helps.
